L-hydroxyproline is a natural component of animal structural proteins such as collagen and elastin. Several microorganisms producing proline trans-4- and cis-3-hydroxylases were identified and these enzymes were used in the industrial production of trans-4- and cis-3-hydroxy-L-proline.

Uses of L-Hydroxyproline.
L-hydroxyproline is a non-essential amino acid, an alpha subamino acid, and a columnar crystal at room temperature. It can be isolated from another amino acid, proline, and is produced by interaction with ascorbic acid vitamin C.

In the food industry, it is often used in combination with additives such as xylitol and sucralose. According to industry data from 2025, its application in the beverage sector accounted for 68%.
Some manufacturers (such as Shandong Zhengfa Biotechnology) also provide industrial-grade products, but food-grade products must meet the national standard GB, with an effective substance content of no less than 98%. Physicochemical Properties of L-Hydroxyproline
L-Hydroxyproline has a water solubility of 36.1% at 25°C, is slightly soluble in ethanol, and exhibits a unique sensory characteristic of bitterness with a distinctive sweetness. Its molecular weight is 131.13, and it belongs to the imino acid class. In collagen, approximately 50% of proline is hydroxylated to 4-hydroxyproline. This substance undergoes a decomposition reaction at 274°C, existing in the solid state as flaky crystals or powder.

Made from gelatin, bone glue, casein, soybean epidermis and other proteins, hydrolyzed with hydrochloric acid, extracted by nitrosation method of sialic acid and then refined and crystallized by resin chromatography. The yield is about 7.0%.
