Extraction Methods

Currently, betulin is primarily obtained through direct extraction, mainly using solvent reflux extraction and recrystallization purification. The specific methods are as follows:

1. Betulin is extracted using an ethanol solution, heated and refluxed for approximately 5 hours. The extract is then distilled under reduced pressure and recrystallized with ethanol 2–3 times to obtain a crude betulin product.

2. Recrystallize the crude product using a methanol/chloroform (1:1) mixture; approximately 300 mL of the methanol-chloroform solution is required for 80 g of crude product. Allow to stand overnight, then filter under vacuum to obtain a white, needle-like solid of betulin.

3. During reflux, the extract was subjected to ultrasonic treatment for 20 minutes. This disrupted the bark tissue, thereby improving the extraction yield and purity of betulin.

4. The optimal conditions for supercritical CO₂ fluid extraction were as follows: modifier dosage of 115 mL/g of birch bark powder; extraction pressure of 20 MPa; extraction temperature of 55°C; and liquid CO₂ flow rate of 10 kg/h.
