The α1-adrenergic receptor is a G protein-coupled receptor: by binding to its natural ligands norepinephrine and epinephrine, it activates phospholipase C and downstream signaling molecules, including inositol triphosphate and diglycerol. Eventually, intracellular calcium levels increase, so smooth muscle contracts. Silodosin is an alpha1-adrenergic receptor antagonist with the highest selectivity for the alpha1a-adrenergic receptor subtype. By blocking the α1a-adrenergic receptor signaling pathway, silodoxin promotes relaxation of the prostate and urethral smooth muscle, thereby improving lower urinary tract symptoms such as urination. Silodoxin also targets the afferent nerves in the bladder, relieving symptoms of bladder overactivity and storage.

Uses of silodosin.

Silodoxin is a selective alpha1A adrenergic receptor antagonist used for the treatment of benign prostatic hyperplasia. Cilodoxine can cause a low probability of transient elevated serum aminotransferase levels and, rarely, clinically significant acute liver injury.
