Carboxymethyl chitosan (CMC) is a carboxymethyl derivative of chitosan. It has strong polarity and is easily soluble in water. It has been widely used in the fields of pharmaceuticals and biomedical material development. Since the content of carboxymethyl chitosan in drugs directly affects their clinical antibacterial and anti-infection effects, it is particularly important to accurately quantify carboxymethyl chitosan.
Uses of Carboxymethyl Chitosan.
Used as high-grade cosmetic additives, heavy metal chelating agents, drug sustained-release agents, plant growth agents, etc.
Preparation of Carboxymethyl Chitosan.
Add sodium hydroxide in isopropanol solution to the reaction flask, the concentration of sodium hydroxide is 40%. Add chitosan with a deacetylation degree of 15% to 45% to the reaction flask, and the mass ratio of chitosan to sodium hydroxide is 1:4.8 after addition. Slowly add the pre-dissolved chloroacetic acid in isopropanone solution to the reaction flask under stirring, and continue stirring and reacting for 3 hours after addition.
The mass ratio of chitosan to chloroacetic acid is 5:1. After the reaction is completed, add concentrated hydrochloric acid to the reacted material, adjust the pH value of the material to 6-7, filter, wash the filter cake 3 times with anhydrous ethanol, and dry the washed filter cake below 50°C to obtain crude carboxymethyl chitosan.
