Preparation of Houttuyfonate Sodium.
(1) Preparation of 2-tridecanone: first add 2.3g of sodium metal in the reaction flask, then add 3mL of anhydrous ethanol, heated to 25-30 ℃, and then add 13.0g of ethyl acetoacetate, 10mL of ethanol and 12.5mL of anhydrous ether solution, stirring and refluxing for 1 hour; and then add 21.85g of lauroyl chloride and 30mL of ether to the reaction flask; cooled and dissolved with dilute sulfuric acid, separated out the ether layer, the aqueous layer was extracted with ether twice and the organic layer was combined and washed with distilled water. Ether was then added to the reaction flask, stirring and refluxing for 2-3 hours; after cooling, it was dissolved with dilute sulfuric acid, and the ether layer was separated, the aqueous layer was extracted twice with ether, the organic layers were combined and washed with distilled water, the ether was recovered, and the intermediate 2-tridecanone was obtained as 15.7g in 92% yield.
(2) the preparation of dodecanoyl acetaldehyde: anhydrous ethanol as a solvent, add 1.1g of sodium metal in the reaction vessel, heating and reflux for 1 hour, dropwise addition of 8g of 2-tridecanone, 3g of ethyl formate, stirring reaction for 4 to 6 hours, leave overnight, and then washed with distilled water, and then extracted with ether for several times, combined with ether solution, dried with anhydrous sodium sulfate and then filtered, the final Finally, the ether was separated, and 8.3g of dodecanoylacetaldehyde was obtained with a yield of 90.3%.
(3) a-hydroxy dodecanoyl ethyl sulfonate sodium preparation: to the reaction vessel to add 60 ml of saturated sodium bisulfite saturated solution, and then slowly add 6.9 g of dodecanoyl acetaldehyde, 85 ~ 90 ° C reaction for 1.5 hours, cool down, filtration, washing, drying, a-hydroxy dodecanoyl ethyl sulfonate sodium 8.1g Yield 89.3%, the content was 99.4% by HPLC.
